Effect of angiogenin overexpression on isolated human meniscus cells

The meniscus is characterized by low blood irrigation because 75% of the meniscus receives
nutrients by passive diffusion through the synovial fluid, reason why the vasculature plays an
essential role in early recovery of a damaged meniscus. Methodology: In patients undergoing
meniscectomy, were isolated and cultured fibrochondrocytes which were transfected with
adenoviral vector Ad-GFP-Ang. The angiogenin level expression as well as the morphological
changes, caused by the overexpression of the latter were evaluated by flow cytometry. Results:
We obtained 90% viability to transfected 4500 MOI of Ad-GFP-Ang, these cells increased the
expression of GFP reporter gene which was considered a di rect marker of protein expression
of angiogenin, which induced phenotypic changes evidenced by expression of surface antigen
CD34 (p < 0.05), and morphological, given by the increase in size and complexity of the
cell. Conclusion: Over expression of angiogenin gene induces phenotypic and morphological
changes that could be associated with early restoration of a meniscus injury.
