Abstract

Mesenchymal Stem Cells (MSCs) has been studied as a suitable source of cells for tissue
engineering and to treat different diseases. At present, Bone Marrow (BM) has been considered the main source to obtained MSC. However, is a painful procedure that requires to be
done in the operating room under anesthesia with their inherent risks and costs. Peripheral
blood (PB) after mobilization is an ambulatory procedure with lower risks that has been considered a novel source of MSC. However, there is no description in the literature about the
isolation of MSC from PB without growth factors administration. Objectives: To isolate MSC
from human peripheral blood of the remnants in the blood bags of conventional donation &
characterize the MSC by flow cytometry. Materials: The remnant of 6 samples of peripheral
blood of adult donors were used. Mononuclear cells were isolated by ficoll gradient, then cells
were cultured in standard conditions during 14 days. Their immunophenotype was assessed
by flow cytometry. Results: In the beginning of culture a population of 8% of the cells were
positive to CD73, CD90 & CD105 markers. However, two different groups of cells were identified: hematopoietic & mesenchymal. At day-14 of culture hematopoietic markers decreased
while mesenchymal markers increased through the time. Conclusions: Is possible to isolate
cells with MSC positive markers from remnants of Human Peripheral Blood with the capability
to multiplicate in standard conditions.
